Semaglutide is a GLP-1 analog with amino acid modifications including Aib substitution at position 8 and C18 fatty acid diacid attachment via glutamate linker. In vitro studies demonstrate high affinity binding to GLP-1 receptors with resistance to DPP-4 degradation.
Animal models show prolonged activation of cAMP signaling pathways due to albumin binding properties. The fatty acid modification significantly extends its half-life while maintaining receptor activation properties.
